Sleeper Cabins are available on some Amtrak routes, usually these are only available on long distance trains where the trip has no transfers. However you can still add a sleeper cabin to reservations with transfers after purchase.
To know if a specific trip has a sleeper cabin option, you will have to select the trip you are interested in, the option for upgrades, including Sleeper cabins will be available on the Trip Summary page.
